Molecular cloning of the B-CAM cell surface glycoprotein of epithelial cancers: a novel member of the immunoglobulin superfamily.

چکیده
The human F8/G253 antigen, B-CAM, is a cell surface glycoprotein that is expressed with restricted distribution pattern in normal fetal and adult tissues, and is up-regulated following malignant transformation in some cell types. We have isolated a complementary DNA for B-CAM using an expression cloning technique. The complementary DNA (EMBL accession number X80026) encodes a 588-amino acid protein which is a novel member of the immunoglobulin superfamily with a characteristic V-V-C2-C2-C2 immunoglobulin domain structure. This structure has been described previously for the human MUC18 melanoma antigen (31% amino acid identity) and chicken and rat versions of a neural adhesion molecule referred to as SC1/DM-GRASP/BEN or KG-CAM, respectively (26% amino acid identity). This homology is suggestive of a role for B-CAM in cell-cell or cell-matrix adhesion. The gene for B-CAM has been mapped by fluorescence in situ hybridization to chromosome 19q13.2-13.3.

Development of specific nanobodies (VHH) for CD19 immuno-targeting of human B-lymphocytes
Objective(s): CD19 is a transmembrane glycoprotein of immunoglobulin superfamily. In order to treat lymphoma, monoclonal antibodies (mAb) can target different antigens, including CD19, CD20 and CD22 on the surface of B-cells. Along with biotechnology progress, a new generation of antibodies is introduced, with the purpose of eliminating the defects of the previous generation. Among the most dev...
متن کاملCRISPR/Cas9, a new approach to successful knockdown of ABCB1/P-glycoprotein and reversal of chemosensitivity in human epithelial ovarian cancer cell line
Objective(s): Multidrug resistance (MDR) is a major obstacle in the successful chemotherapy of ovarian cancer. Inhibition of P-glycoprotein (P-gp), a member of ATP-binding cassette (ABC) transporters, is a well-known strategy to overcome MDR in cancer. The aim of this study was to investigate the efficiency and ability of CRISPR/Cas9 genome editing technology to knockdown ABCB1 gene expression ...
متن کاملDevelopment of a Stable Cell Line, Overexpressing Human T-cell Immunoglobulin Mucin 1
Background: Recent researches have demonstrated that human T-cell immunoglobulin mucin 1 (TIM-1) glycoprotein plays important roles in regulation of autoimmune and allergic diseases, as well as in tumor immunity and response to viral infections. Therefore, targeting TIM-1 could be a potential therapeutic approach against such diseases. Objectives: In this study, we aimed to express TIM-1 protein...
متن کاملcDCC (chicken homologue to a gene deleted in colorectal carcinoma) is an epithelial adhesion molecule expressed in the basal cells and involved in epithelial-mesenchymal interaction.
Cloning of human DCC (deleted in colorectal carcinoma, Fearon et al., 1990) showed that it is an immunoglobulin superfamily member homologous to neural cell adhesion molecules (N-CAM). To explore the normal function of this molecule, we have cloned a chicken homologue to DCC (cDCC) and raised an antibody to DCC. cDCC is a protein of 160 kDa with an expression pattern distinct from those of othe...
متن کاملPreparation and characterization of a novel nanobody against T-cell immunoglobulin and mucin-3 (TIM-3)
Objective(s): As T-cell immunoglobulin and mucin domain 3 (TIM-3) is an immune regulatory molecule; its blocking or stimulating could alter the pattern of immune response towards a desired condition. Based on the unique features of nanobodies, we aimed to construct an anti-TIM-3 nanobody as an appropriate tool for manipulating immune responses for future therapeutic purposes. Materials and Meth...
